چکیده

Purine nucleoside phosphorylase (PNP) is an ubiquitous enzyme which plays an important role in arsenic (As) detoxification. As is a toxic metalloid present in air, soil and water; is abundant in the environment and is readily transferred along the trophic chain, being found even in human breast milk. Milk is the main nutrient source for the growth and development of neonates. Information on breast milk synthesis and its potential defense mechanism against As toxicity is scarce. In this study, PNP and antioxidant enzymes activities, as well as glutathione (GSH) and total arsenic (TAs) concentrations, were quantified in breast milk samples. PNP, superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST), glutathione peroxidase (GPx), glutathione reductase (GR) activities and GSH concentration were determined spectrophotometrically; TAs concentration ([TAs]) was measured by atomic absorption spectrometry. Data suggest an increase in PNP activity (median = 0.034 U mg protein-1) in the presence of TAs (median = 1.16 g L(-1)). To explain the possible association of PNP activity in breast milk with the activity of the antioxidant enzymes as well as with GSH and TAs concentrations, generalized linear models were built. In the adjusted model, GPx and GR activities showed a statistically significant (p<0.01) association with PNP activity. These results may suggest that PNP activity increases in the presence of TAs as part of the detoxification mechanism in breast milk.

IL-8 May Be the Target of Arsenic in Human Breast Milk

Background and Aims: The critical aim of this study was to evaluate the effects of breast-milk contamination with Nickel and Arsenic, as well as aflatoxin M1 (AFM1) on the milk levels of IL-6 and IL-8.  Materials and Methods: Breast milk was collected from 76 mothers on the 30th day post-parturition and milk levels of AFM1, Nickel, Arsenic, IL-6 and IL-8 were evaluated. Results: The results s...

Purine Nucleoside Phosphorylase Targeted by Annexin V to Breast Cancer Vasculature for Enzyme Prodrug Therapy

BACKGROUND AND PURPOSE The targeting of therapeutics is a promising approach for the development of new cancer treatments that seek to reduce the devastating side effects caused by the systemic administration of current drugs. This study evaluates a fusion protein developed as an enzyme prodrug therapy targeted to the tumor vasculature. Cytotoxicity would be localized to the site of the tumor u...

Alcohol and Lactation

Background: Breastfeeding is the safest and best method for nurturing infant growth and health. While the harmful effects of alcohol during pregnancy are well-established, the consequences of alcohol intake during lactation have been far less examined. The effects of alcohol intake with large amounts noted on the infant include drowsiness, diaphoresis, deep sleep, weakness, decrease in linear g...

Expression of purine metabolism-related enzymes by microglial cells in the developing rat brain.

The nucleoside triphosphatase (NTPase), nucleoside diphosphatase (NDPase), 5'-nucleotidase (5'-Nase), and purine nucleoside phosphorylase (PNPase) activity has been examined in the cerebral cortex, subcortical white matter, and hippocampus from embryonic day (E)16 to postnatal day (P)18. Microglia display all four purine-related enzymatic activities, but the expression of these enzymatic activi...

An Assessment of Oxidative Stress and Antioxidant System Activity in Alfalfa Plants Treated with Different Forms of Mineral Arsenic

Arsenic (As), a metalloid and unnecessary element, is highly toxic to living organisms. It can be accumulated in the environment and enter the food chains through uptake by plants. The present study was designed to assess the response of antioxidant defense system of alfalfa to As (As0, As3+ and As5+) induced oxidative stress at four rates (0, 100, 200, and 300 µM) as a completely randomized de...
